Stochastic HIV model coupled with pharmacokinetics and drug adherence may explain intermittent viral blips

چکیده
We develop a stochastic HIV model by integrating drug adherence into pharmacokinetic and coupling pharmacodynamic with viral dynamic model. Numerical simulations show that the proposed can generate blips, which have been observed in patients receiving suppressive antiretroviral therapy. calculate probability density function of infected CD4+ T cells developing generalized evolution equation. fit to clinical data four exhibiting blips. The results demonstrate poor be reason explaining occurrence blips treated patients. also find dynamics are sensitive drug-adherence parameters, significant impact on frequency amplitude modeling methods applied study long-term therapy other chronic diseases might an issue.

Intermittent HIV-1 viremia (Blips) and drug resistance in patients receiving HAART.
CONTEXT Many patients infected with human immunodeficiency virus type 1 (HIV-1) and receiving highly active antiretroviral therapy experience intermittent episodes of detectable viremia ("blips"), which may raise concerns about drug resistance, lead to costly repeat measurements of viral RNA, and sometimes trigger alterations in therapy. OBJECTIVE To test the hypothesis that blips represent r...
متن کاملRole of viral load blips, drug boosting, and therapeutic holidays in HIV patient management.
IAS is a nonpro f i t organization founded in 1988, with more than 10,000 members representing more than 130 countries. This article addresses three main concepts discussed at the meeting that appear to play a role in the way clini-cians and laboratorians are currently approaching HIV patient management. Viral load blips are temporary spikes in viral load above 50 copies/mL that go back to unde...
متن کاملSocioeconomic factors explain suboptimal adherence to antiretroviral therapy among HIV-infected Australian adults with viral suppression
BACKGROUND Missing more than one tablet of contemporary antiretroviral therapy (ART) per month increases the risk of virological failure. Recent studies evaluating a comprehensive range of potential risk factors for suboptimal adherence are not available for high-income settings. METHODS Adults on ART with undetectable viral load (UDVL) were recruited into a national, multi-centre cohort, com...

متن کاملa stochastic model for the ethanol pharmacokinetics
background: the aim of this study was to propose a new stochastic model to study the time course of ethanol elimination in human bodies. methods: the times and amount of alcohol ingested are assumed to be random in controllable intervals. constant elimination rate follows zero order kinetics and is replaced by first order kinetics when the effects of alcohol increase due to alcohol ingestion. s...
